Usuthu are coming into the encounter undefeated in their last two matches including their first-ever win in the Champions League stage when they defeated Guinea's Horoya AC 1-0 last week Friday.
The KwaZulu-Natal giants are currently placed third on the group standings - level on points with second-placed Setif and three points behind leaders, Raja Casablanca of Morocco.
Usuthu were held to a 0-0 draw by a stubborn TS Galaxy side in their DStv Premiership clash on Tuesday and their coach Benni McCarthy has urged his charges to focus on their game against Setif.
“We let a big one slip, this was a game where we probably could’ve gotten easier three points more than any other game," McCarthy said.
"But it was not to be and now we can focus and concentrate on Friday on the CAF Champions League."
Meanwhile, Setif have been inconsistent having recorded their second defeat in their last three competitive matches when they succumbed to a 1-0 loss to Raja in a Group B game last week Friday.
Despite the home defeat, the Algerian giants are placed second on the standings having started their Group B campaign with a 1-0 win over Horoya at home almost two weeks ago.
Nabil Kouki, the Setif head coach, was left to rue his side's lack of efficiency following their clash with three-time African champions Raja and he will be hoping for an improved display against AmaZulu.
"We have two or three chances that we didn't know how to exploit," Kouki, who guided Setif to a second spot finish in the Algerian Ligue Professionnelle 1 last season, said.
"We conceded following a defensive error and we could not come back to the score. We faced a team like Raja, very experienced, we lacked efficiency."
