Johnny Ferreira's side qualified for the competition proper after beating Pretoria Callies 3-1 in the preliminary round and they eliminated ABC Motsepe League outfit African All Stars by the same scoreline in the round of 32.
Venda aren't in the best form coming into this cup-tie, with the 1-1 draw against JDR Stars on Sunday extending their winless run to three outings.
However, their debut season in the second tier is going well, having lost just four of their 21 matches to sit sixth in the table, five points adrift of leaders Richards Bay.
Meanwhile, AmaTuks recorded a 2-1 victory over Hungry Lions in the preliminary round to set up a round-of-32 date with DStv Premiership club Chippa United.
Tlisane Motaung's men scored twice in the final 10 minutes to secure a 2-0 win against the Chilli Boys and they remain unbeaten since claiming that top-flight scalp.
A 2-0 success over Platinum City Rovers on Sunday made it three wins from four in the league as Tuks closed within three points of the summit in third place.
University of Pretoria's record in the tournament hasn't been great since they reached the final all the way back in 2009, where they lost 1-0 to Swallows.
They made it to the quarterfinals the following year, but haven't gone further than the round of 16 since then.
The two teams could not be separated when they met at the same venue on November 28 as Tebogo Mohlamonyane (13') quickly cancelled out Rankoko Tau's ninth-minute opener for Venda to earn a 1-1 draw.
