The action gets under way on Friday with Cape Town Spurs hosting Polokwane City at Athlone Stadium and NB La Masia welcoming Venda Football Academy to the Bidvest Stadium.
The 12th-placed Urban Warriors are aiming to snap a three-game winless run as they seek a first victory over Rise And Shine since September 2017.
The Limpopo outfit recorded their third win in four outings when they beat Hungry Lions 2-0 on Sunday to move up to third in the standings.
La Masia (13th) suffered their third defeat in four matches after losing 2-1 to Pretoria Callies last weekend, with Venda moving up to 12th spot after a 1-0 win over Baroka arrested a run of two successive losses.
TTM will attempt to regain top spot for at least a day when they take on Baroka at the Global Stadium on Saturday.
Second-placed Vhadau Vha Damani were pushed off the summit following a 1-1 draw against Magesi, which halted a run of three consecutive wins. Bakgaga (9th) saw a three-match unbeaten run come to an end with a 1-0 reverse to Venda last time out.
Elsewhere, Platinum City Rovers will be hoping to get their campaign up and running when they lock horns with fellow strugglers Black Leopards at the Malamulele Stadium.
Rock-bottom Rovers managed to stop the rot with a 1-1 draw against the Urban Warriors on Monday, although they remain winless in five games. Lidoda Duvha dropped into the relegation zone after going down 3-1 to Casric Stars - their fourth defeat of the season.
Casric Stars will be eyeing back-to-back wins when Pretoria Callies visit the Solomon Mahlangu Stadium, while JDR Stars and Hungry Lions square off at the Giant Stadium.
The 14th-placed Romans will be hoping to build on their first victory of the campaign as they attempt to put some more distance between themselves and the drop zone.
Meanwhile, the Hammer Boys (8th) lost ground on the frontrunners after a 2-0 defeat to AmaTuks halted a three-match unbeaten run and the Lions dropped to sixth spot after losing 2-0 to Polokwane City on Sunday.
Round 6 concludes on Sunday with University of Pretoria entertaining All Stars at the Tuks Stadium, while Uthongathi do battle with Magesi at the Princess Magogo Stadium.
The league leaders are bidding to make it four wins in a row as they seek to end All Stars' unbeaten start to proceedings.
The Cane Cutters find themselves two points off the summit in fourth place following back-to-back draws against La Masia (1-1) and All Stars (0-0), with Magesi (10th) holding TTM to a 1-1 stalemate most recently.
