The 55-year-old Australian has been with the London team in different roles since 2018 and was in charge for their 2021 Premiership title triumph.
"This decision is not one that's been made lightly but I'm in a position to take on a new challenge in my rugby career, while spending time on a family project," Millard said.
Quins are currently fourth in the Premiership table with six games of the regular season remaining.
The announcement of Millard's departure from The Stoop comes after he was linked to the vacant post of Wales' director of rugby, as the men's team looks to end a 17-test losing streak – the longest for a Tier-One nation in the professional era.
