The 31-year-old uncapped Northampton tighthead replaces Leicester prop Will Hurd, sidelined by a foot injury.
Meanwhile fly-half Adam Hastings, the son of Scotland great Gavin Hastings, has also been ruled out by a knee problem.
Millar-Mills, born in England, qualifies for Scotland through his mother.
His sisters are both internationals, with Bridget playing for Scotland and Harriet capped by England.
Millar-Mills joined his teammates on Monday.
Scottish Rugby also said Tuesday that Javan Sebastian and Jamie Dobie will train with the squad as they recover from injury.
Scotland, knocked out in the pool phase of last year's World Cup in France, begin their Six Nations campaign away to Wales in Cardiff on February 3.
The Scots will be bidding for their first win in the Welsh capital since 2002.
